Biodegradation of graphdiyne oxide in classically activated (M1) macrophages modulates cytokine production

نویسندگان

چکیده

Human macrophages are reprogrammed by graphdiyne oxide from M2 to M1 macrophages, leading degradation of GDYO through a peroxynitrite-driven pathway with subsequent modulation pro-inflammatory cytokines including IL-1?.

Arginase modulates nitric oxide production in activated macrophages.

In macrophages and many other cell types,l-arginine is used as a substrate by both nitric oxide synthase (NOS) and arginase to produce nitric oxide (NO) and urea, respectively. Because the availability ofl-arginine is a major determinant for NO synthesis in the activated macrophage, we hypothesized that NO production may be reduced by arginase via depleting the common substrate in this cell typ...

Elimination of classically-activated macrophages in tumor-conditioned medium by alternatively-activated macrophages

Cellular interactions are critical during development, tissue fitness and epithelial tumor development. The expression levels of specific genes confer to tumoral cells a survival advantage versus the normal neighboring cells. As a consequence, cells surrounding tumors are eliminated and engulfed by macrophages. We propose a novel scenario in which circulating cells facing a tumor can reproduce ...

Apoptotic Cell Effects on Nitric Oxide Production in Activated Macrophages

Macrophages can be activated to kill tumor necrosis factor (TNF)-sensitive, nitric oxide (NO)resistant 3T3-VSV and NO-sensitive, TNF-resistant P815 cells. Activated macrophages co-cultivated with 3T3-VSV but not P815 targets show decreased production of NO. Since TNF can induce apoptosis and apoptotic cells can interfere with macrophage activation, apoptotic cells were compared to normal cells ...

Effect of Neutrophils on Nitric Oxide Production from Stimulated Macrophages

Background: During the initial phase of an infection, there is an upregulation of inducible nitric oxide synthase in the macrophages for the production of nitric oxide. This is followed by the recruitment of polymorphonuclear leukocytes (neutrophils) which release arginase. Arginase competes with inducible nitric oxide synthase for a common substrate L-arginine. Objective: To investigate whethe...
